🫘 Miso, soy and koji: East Asia's mould
You open a bag of rice koji and it smells of chestnuts. The yellow felt is a domesticated mould  and the reason miso and soy sauce exist.
What you’ll learn
- The bag that smells of chestnutsName koji as cooked grain grown with Aspergillus oryzae for about 48 hours, and treat the yellow felt as a living enzyme source, not a sauce.A. oryzae on rice, barley or soy. Spores, steam, two days. The paste comes later.
- A factory of enzymesUse Machida's expanded hydrolases and Ito's three glutamate doors to explain why koji, not the later vat, does the first demolition.37 Mb, extra secretory enzymes. Amylase, protease, glutaminase. Amazake is the sugar fork.
- The cousin that makes poisonPlace A. oryzae inside A. flavus group I, and treat aflatoxin safety as a ruined or half-deleted gene cluster plus selected metabolism, not as folklore.Geiser's clade. Tominaga's ruin. Gibbons's cell factory. Tradition kept the strains; the genome is why you can eat them.
- Jiang before misoFollow Shurtleff's documents from Zhouli jiang through Qimin Yaoshu doujiang to Japanese hishio and the ninth-century word miso, without a single-inventor story.Meat jiang first. Soybean method in 533–544. Jiangyou is pressed jiang. Korea kept meju.
- The mash you press and the mash you keepSeparate koji from the brine relay of Z. rouxii and T. halophilus, and treat miso versus shoyu as keep-or-press, with colour as time and rice ratio.48 hours, then months. White is short rice. Red is long soy. Shoyu is the juice.
- What you can still taste tonightTaste pale against dark miso, keep tempeh and natto off the koji list, and leave the domestication birthday unsolved.Doenjang, doubanjiang, a soup bowl. The enzymes are dated. The birthday is not.
Questions this course answers
What is koji, in the sense this course uses the word?
Shurtleff: steamed grain plus A. oryzae, about 48 hours. The paste and the sauce come later.
Match each koji tool to the job Ito and Machida actually give it
Ito 2021: hydrolases feed the vat. Machida 2005: the genome expanded those secretory enzymes.
Put these in the order the documents actually allow
Shurtleff: Zhouli third century BC; Qimin Yaoshu 533–544; Man'yoshu 686; miso 886–901.
What did Tominaga find in the aflatoxin-cluster homolog of A. oryzae?
Tominaga 2006: group 2 (36.7%) keep only vbs, verB and avnA in half the cluster. Even group 1 does not express the aflatoxin genes.
About how large is the A. oryzae RIB 40 genome Machida published in 2005?
Machida 2005: 37 Mb, 12,074 genes, 7–9 Mb larger than A. nidulans and A. fumigatus.
What is the honest difference between traditional miso and honjozo soy sauce?
Shurtleff's two-part miso; Ito's honjozo moromi then a press. Same mould idea, last gesture differs.
